At this time, since the steam cannot overflow, the pressure in the sterilization pot is increased, thereby increasing the boiling point. Temperatures above 100°C were obtained. It leads to the coagulation and denaturation of bacterial protein to achieve the purpose of sterilization. When using the pressure steam sterilizer for sterilization, it is extremely important to remove the cold air in the sterilizer, because the expansion pressure of air is greater than the expansion pressure of water vapor, so when the water vapor contains air, under the same pressure, the The temperature of air vapor is lower than that of saturated vapor. So the sterilization of saturated steam is more thorough!

1. Water should be added to the water level line before the autoclave is used;

Put the culture medium, distilled water or other utensils to be sterilized into the pressure steam sterilizer, close the lid, and check the status of the exhaust valve and safety valve (the upper and lower exhaust valves are opened and the safety valve is closed and then heated to facilitate air discharge. Both the upper and lower exhaust valves are in the open state at the same time until water vapor overflows from the upper and lower valves. After this state is maintained for 7 or 8 minutes, the upper exhaust valve is closed after the air is exhausted, and the lower exhaust valve is reduced to ensure safety. valve remains closed);

3. Turn on the power, check whether the parameter settings are correct, and then press the "work" button, the sterilizer starts to work; the air conditioner is automatically sent, and when the temperature reaches 105 °C, the bottom exhaust valve is automatically closed, and then the pressure begins to rise;

4. When the pressure rises to 0.15MPa (121°C), the autoclave will automatically deflate again, and then start to count the time. Generally, the medium is sterilized for 20 minutes, and the distilled water is sterilized for 30 minutes;

5. After reaching the specified sterilization time, turn off the power supply and open the vent valve to slowly release the gas; when the pressure pointer drops to 0.00MPa, the lid can only be opened when there is no steam discharged from the vent valve. .
